Good day.
2 questions....
Let say I have an image and there a 2 circles on the image and I need to align these for exact measurements in Moi...is this possible?
kind of like scaling based on 2 points?
Also I have Moi4.0 and running Moi5.0 but it seems I change the shortcut but when I am going to use it it says plugin not found or If I setup a shortcut..when it does not work...check it..it changed to the old one.

Yes, there's an "Align" tool in the View > Image command that will do that.

MoI will save shortcuts to the moi.ini file when it exits.
So when you set up a new shortcut don't be running 2 instances of MOI at that same time. Make sure you only have one window running and close it before starting up multiple instances.
For "plugin not found", where are you copying the plugin to? Put it in the appdata commands folder so it can be found by all versions. Don't put it in the commands folder inside of c:\Program Files.
